Fluorescence microscopy images show exosomes loaded using Exo-Fect™ with either a Texas Red end-labeled siRNA (left panels) or unlabeled siRNA (right panels). Following loading, exosomes were captured on Exo-Flow™ CD63 magnetic beads and immobilized for imaging, demonstrating successful incorporation of siRNA cargo into exosomes.

Exosomes loaded with siRNA using Exo-Fect™ (as shown in Figure 1) were eluted from Exo-Flow™ CD63 magnetic beads and subsequently incubated with HEK293 cells. Fluorescence detected in recipient cells confirms the successful delivery and cellular uptake of the Texas Red–labeled siRNA cargo mediated by Exo-Fect™–transfected exosomes.

Exo-Fect™ is compatible with larger nucleic acid cargos, including mRNA and plasmid DNA. (Left panels) Exosomes were loaded with 1 µg of mRNA encoding red fluorescent protein (RFP) using Exo-Fect™ and subsequently incubated with HEK293 cells. RFP expression was detected 24 hours post-treatment, indicating successful mRNA delivery and translation. (Right panels) Exosomes were loaded with 5 µg of GFP-encoding plasmid DNA using Exo-Fect™ and added to HEK293 cells. GFP expression was observed 48 hours post-treatment, confirming effective plasmid delivery and gene expression. The presence of RFP and GFP fluorescence in recipient cells demonstrates efficient cargo transfer mediated by Exo-Fect™-loaded exosomes.

Exosomes were loaded with the small molecule cumate using Exo-Fect™ and subsequently incubated with cells harboring a cumate-inducible GFP reporter construct. GFP expression was observed only in cells treated with cumate-loaded exosomes, demonstrating successful encapsulation and delivery of small-molecule cargo mediated by Exo-Fect™.
